They released a short statement after the club suffered a 0-2 loss at home to Bristol Rovers which leaves Orient sitting 18th in the table.
The Sky Bet League 2 club haven’t won a game since they beat Swindon Town 4-1 back on December 7.
A run of ten league games in which they have scored just two goals.
Jackett’s assistant, Joe Gallen, has also departed the club.
The club’s statement thanked both Jackett and Gallen and wish them well for the future.
Jackett joined the club in the summer after a four year stint at Portsmouth.
He leaves with a win percentage of 31.6% from the 38 games he took charge of.
