Eight fire engines and around 60 firefighters station crews were called to the fire on Hoe Street.
A single-storey building at the back of a shop on the road was completely destroyed by the blaze.
Three men were rescued from the building by fire crews wearing breathing apparatus.
The men were treated at the scene by London Ambulance Service and the Helicopter Emergency Medical Service before being taken to hospital.
London Fire Brigade was called at 5.58am on the morning of Thursday, 8 April and the fire was brought under control at 8.20am.
Fire crews from Walthamstow, Leyton, Tottenham, Woodford, Homerton and Bethnal Green fire stations attended the scene.
The cause of the fire will be investigated by the London Fire Brigade and the Metropolitan Police Service.
