The illegal event was discovered in a disused shop, in Green Street on Saturday night.
Officers were made aware of the gathering shortly after 10.30pm.
Police attended and gained entry before dispersing the crowd.
The event was finally closed by 3am on Sunday, 25 April.
Officers remained in the area after and a dispersal zone was implemented in the vicinity for a 48-hour period.
The organiser was reported for consideration of a £10,000 fixed penalty notice.
Inspector Garside, from the North East BCU, which covers Newham, said: “Despite the current easing of lockdown restrictions, we remain in a pandemic and this type of large gathering remains prohibited.
“Not only were those present taking unnecessary risks, they were also causing significant disruption to the local area.
“The behaviour of a minority will not be tolerated and we will continue to do whatever we can to keep our communities safe.”
